How to Read HPLC & Mass Spectrometry Certificates of Analysis

Reversed-phase HPLC (RP-HPLC) separates a peptide sample into its component peaks on a C18 column. The area percentage of the principal peak represents the compound's purity. A single dominant sharp peak with area ≥98.0% indicates a high-purity compound. Multiple peaks indicate impurities or degradation products.

Mass spectrometry (MS) confirms molecular identity by measuring the mass-to-charge ratio (m/z) of the ionised peptide. The observed molecular mass is compared against the theoretical mass calculated from the amino acid sequence.
