HPLC and Mass Spectrometry Explained: The Gold Standard in Peptide Analysis

High-Performance Liquid Chromatography (HPLC) is the foundational analytical technique for determining the purity of synthesized peptides in the UK research market.

Unlike simple mass spectrometry, which only confirms the molecular weight of the compound, HPLC physically separates the target peptide from synthesis impurities (like truncated sequences or deletion errors). When evaluating a Certificate of Analysis (COA), researchers should always request the raw HPLC chromatogram. A single, sharp peak indicates high purity. Multiple peaks or broad, jagged peaks indicate the presence of impurities that could completely skew in-vitro assay results.

Mass Spectrometry (MS) is then paired with HPLC to confirm the structural identity of that single peak. If the mass-to-charge ratio matches the theoretical weight of the peptide (e.g., 4111 g/mol for Tirzepatide), identity is confirmed.
